MiR-493-5p alleviates lipopolysaccharide-induced inflammation in ATDC5 chondrogenic cells by targeting TAB2

Purpose: To investigate the effect of miR-493-5p in lipopolysaccharide (LPS) -induced ATDC5 chondrogenic cells.
 Methods: The MTT assay was used to determine viability LPS-induced cells. ENCORI (starbase.sysu.edu.cn/) predict target miR-493-5p. Quantitative reverse-transcription-polymerase chain reaction (qRT-PCR) expression controlled cells and treated with various combinations LPS, negative control miRNA (NC), mimic, TAB2 overexpression vector, inhibitor. qRT-PCR, while western blot assess mRNA protein levels control, LPS + NC, inhibitor groups. qRT-PCR ELISA were evaluate TNF-α, IL-18, IL-1β expressions mimic. Furthermore, two techniques also inhibitor, overexpression, mimic groups.
 Results: MiR-493-5p downregulated (p < 0.001). Overexpression increased cell but decreased inflammatory factors targeted inhibited reversed suppression by injury 0.001).
 Conclusion: alleviates inflammation targeting TAB2. Thus, have potentials for use as clinical therapeutic targets OA.
